You can tranfer players at any time. You get one free tranfer each gameweek, after that they cost 4 points each. When you sell a player, there's a sell on fee of half your profit. If you buy a player for 7, his price goes up to 8 (The more people transfer in a player, the higher his value goes and vice versa) then you can sell him for 7.5.
